Mysterious coolant loss ? - Tifosi
ya. every once in a wile. it never smokes, and the coolent level is consistant than one day its down half an inch. did u have same problem? searched forums for single cylinder intermittent misfires and didnt find any solutions. just people saying coils, plugs, ICM, and injectors, but i checked all that!! please help!

have you examined the plug for that cylinder? was the tip white? have you swapped coil packs?
have you changed that plug? (could be fouled)
i'd be *way* more concerned about idetifying the source of the coolant loss than a misfire issue. if you have eliminated all external coolant loss possibilities then only one other option remains - head gasket.
examine the plug. if it's white then it could be coolant entering via bad head gasket (though i don't know that misfiring is a symptom)
get an oil analysis to determine if oil is contaminated with coolant via headgasket.

order an oil sample kit from them then do an oil change and take a sample. send it to them and they will analyze for about $20.

From your description, I agree with berTTos and thinks it may be a head gasket leak....

i had a small leak coming from my coolant temp sensor - small crack in the housing
and coolant was slowly overflowing.
Same symptoms, consistent level most of the time but then one day I was down quite a bit.

1. Pressurize cooling system with the engine off and visually look for leaks .........
around the after-run pump, coolant temp. sensor, waterpump, etc.
(I had a bad o-ring on the coolant temp. sensor)
2. Do a Blackstone Oil Analysis to see if there's any coolant in your engine oil.
